Induction of Protection against Paraquat-induced Oxidative Damage by Abscisic Acid in Maize Leaves is Mediated through Mitogen-activated Protein Kinase被引量:4

Mitogen-activated protein kinase (MAPK) cascade has been shown to be important components in stress signal trans-duction pathway. In the present study, protection of maize seedlings (Zea mays L.) against paraquat-generated oxidative toxicity by abscisic acid (ABA), its association with MAPK and ZmMPK5, a candidate for MAPK were investigated. Treatment of maize leaves with exogenous ABA led to significant decreases in the content of malondialdehyde, the percentage of ion leakage and the level of protein oxidation (in terms of carbonyl groups) under paraquat (PQ) stress. However, such decreases were blocked by the pretreatment with two MAPK kinase inhibitors PD98059 and U0126. The damage caused by PQ was further aggravated by inhibitors. Two inhibitors also suppressed the total activities of the antioxidant enzymes superoxide dismutase (SOD, EC 1.15.1.1), catalase (CAT, EC 1.11.1.6), ascorbate peroxidase (APX, EC 1.11.1.11), and glutathione reductase (GR, EC 1.6.4.2). Besides, treatment with PQ stimulated the activation of a 46 kDa MAPK, which was identified as ZmMPK5 by in-gel kinase assay with immunoprecipitation. These results reveal that ABA-induced protection against PQ-generated oxidative damage is mediated through MAPK cascade in maize leaves, in which ZmMPK5, a candidate for MAPK, is demonstrated to be involved.

水分胁迫诱导玉米Zm rboh基因表达及ABA在其中的作用被引量:12

利用定量和半定量PCR方法检测了玉米野生型及ABA缺失突变体vp5的ZmrbohA/B/C/D基因在水分胁迫下的表达情况。结果发现,水分胁迫能诱导玉米野生型ZmrbohA/B/C/D基因表达,并且表达在水分胁迫处理60 min、150 min时分别达到2个高峰,渗透势为-0.7 MPa的水分胁迫诱导效果尤其显著;同样处理下vp5的ZmrbohA/B/C/D基因表达没有明显变化。表明:ABA参与了水分胁迫诱导ZmrbohA/B/C/D基因表达的过程。

一氧化氮在脱落酸诱导的玉米叶片亚细胞抗氧化防护中的作用(英文)被引量:1

研究了ABA诱导NO产生的来源以及NO在ABA诱导的玉米叶片H_2O_2累积和亚细胞水平抗氧化中的作用。ABA诱导玉米叶片NO的产生以及NOS活性增加,NOS抑制剂抑制这种增加。NO清除剂和NR抑制剂预处理也抑制了ABA诱导的NO产生,但是并不影响ABA诱导的NOS活性,结果提示了ABA诱导的NO的产生来源于NOS和NR 2条途径。NO清除剂、NOS抑制剂和NR抑制剂预处理抑制了ABA和H_2O_2诱导的抗氧化防护酶基因SDD4、cAPX、GR1的表达和叶绿体及细胞溶质抗氧化酶活性的增加,表明NO参与ABA和H_2O_2诱导的玉米亚细胞抗氧化防护系统。另一方面,以NO供体SNP预处理减少了ABA诱导的H_2O_2的累积,而c-PTIO逆转了SNP减少ABA诱导的H_2O_2累积的作用。SNP处理诱导了亚细胞抗氧化酶活性的增加,用c-PTIO预处理抑制了这种增加。实验结果表明ABA诱导H_2O_2和NO产生,NO上调了玉米亚细胞抗氧化防护酶活性,进而防止玉米叶片中H_2O_2的过量累积。因此在玉米ABA诱导的信号转导中有一个NO和H_2O_2负反馈环。

Alternative Splicing and Differential Expression of Two Transcripts of Nicotine Adenine Dinucleotide Phosphate Oxidase B Gene from Zea mays被引量:6

With the exception of rice,little is known about the existence of respiratory burst oxidase homolog(rboh) gene in cereals. The present study reports the cloning and analysis of a novel rboh gene,termed ZmrbohB,from maize(Zea mays L.) . The full-length cDNA of ZmrbohB encodes a 942 amino acid protein containing all of the respiratory burst oxidase homolog catalytically critical motifs. Alternative splicing of ZmrbohB has generated two transcript isoforms,ZmrbohB-α and-β. Spliced transcript ZmrbohB-β retains an unspliced intron 11 that carries a premature termination codon and probably leads to nonsense-mediated mRNA decay. Expression analysis showed that two splice isoforms were differentially expressed in various tissues and at different developmental stages,and the major product was ZmrbohB-α. The transcripts of ZmrbohB-α accumulated markedly when the maize seedlings were subjected to various abiotic stimuli,such as wounding,cold(4°C),heat(40°C),UV and salinity stress. In addition,several abiotic stimuli also affected the alternative splicing pattern of ZmrbohB except wounding. These results provide new insight into roles in the expression regulation of plant rboh genes and suggest that ZmrbohB gene may play a role in response to environmental stresses.

水分胁迫下玉米叶片中一氧化氮与钙和钙调素的相互作用被引量:4

以玉米(Zea mays L.)为材料,研究水分胁迫下玉米叶片中NO与Ca2+和钙调素(CaM)含量之间的关系。结果显示:Ca2+鳌合剂、Ca2+通道阻塞剂和CaM拮抗剂预处理均抑制了水分胁迫诱导的NO产生和一氧化氮合酶(nitric ox-idesynthase,NOS)活性的提高,同时也能抑制水分胁迫诱导的叶片线粒体NO产生,而对叶绿体NO产生并没有影响;NO清除剂羟基2苯基4,4,5,5四咪唑1羟基3氧化物(2-4-carboxypheny l-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide,c-PTIO)能抑制水分胁迫条件下Ca2+和CaM的产生以及CaM1基因表达,但对水分胁迫最初诱导的玉米叶片叶肉细胞原生质体Ca2+和玉米叶片中CaM1基因表达以及CaM含量增加没有影响。上述结果表明:水分胁迫下NO和Ca2+/CaM之间存在相互作用。
