猪胸膜肺炎放线杆菌外膜蛋白基因(OMP-1)的克隆、表达及其ELISA方法的建立被引量:2

猪传染性胸膜肺炎由猪胸膜肺炎放线杆菌(APP)引起,是猪最重要的呼吸道传染病之一。通过筛选APP 3~8 kb随机基因组文库,获得1株阳性克隆。测序拯救质粒得到其DNA序列,通过BLAST分析确定此片段为APP外膜蛋白基因,预计成熟蛋白质的分子量为48.766 kD。根据序列设计特异性引物,用PCR扩增该基因,分子克隆表达该外膜蛋白。Western blot结果表明,该蛋白与APP康复期血清发生反应。纯化该重组蛋白,并将其作为抗原包被ELISA酶标板,对已知阳性血清、阴性血清和临床送检血清样品进行ELISA检测,为最终建立APP ELISA检测方法奠定了基础。

犬源粪肠球菌的分离与鉴定被引量:1

随着都市宠物业的发展,伴侣动物的化脓性疾病日趋增多。为探究其影响病原,本研究对一例犬子宫蓄脓的浓汁进行了病原学鉴定。利用细菌学检测方法对采集到的病料进行分离纯化、涂片染色、镜检和生化鉴定,然后采用16S r RNA基因序列分析鉴定,发现该分离株的形态特征、生化结果与粪肠球菌一致,分离菌株的16S r RNA也与参考的粪肠球菌菌株同源性高达99%。药敏试验检测发现,该菌株对四环素、青霉素G、米诺环素、红霉素、诺氟沙星、环丙沙星、万古霉素、左氟沙星、克林霉素、多粘菌素B、氯霉素、复方新诺明和克拉霉素表现为耐药,对苯唑西林和呋喃妥因敏感。这些研究结果可为治疗此类多重耐药性肠球菌感染提供参考。

犬源铜绿假单胞菌的分离与鉴定被引量:3

为了确定一株从犬子宫蓄脓的脓汁中分离的细菌的属性及致病性和敏感药物,试验采用病原分离培养、生化试验、动物致病性试验、药敏试验及16S rRNA基因鉴定方法对该菌株进行研究。结果表明:分离菌为铜绿假单胞菌,具有较强的致病性,对环丙沙星、氧氟沙星、头孢类抗生素敏感,对其他抗菌药物如青霉素、阿奇霉素、四环素、万古霉素等耐药。

副猪嗜血杆菌免疫机制研究进展被引量:1

副猪嗜血杆菌(Hps)是常常存在于猪上呼吸道的革兰氏阴性菌,也是革拉泽氏病的病因,该病是以多发性浆膜炎、关节炎和脑炎为特征的全身炎症感染。近年来,对Hps的天然免疫研究表明,猪肺泡巨噬细胞(PAMs)能够差异地上调与炎症和吞噬有关的多种基因,细胞在接触Hps时会产生多种促炎细胞因子。Hps菌株对PAMs吞噬的易感性和补体的杀菌作用受毒株的毒力表型影响。然而,在有Hps特异性抗体的情况下,强毒株容易被吞噬。抗菌药物在养猪业中被普遍用于治疗和控制革拉泽氏病,一些常见的抗菌药物已被证明可以减少Hps的定植。本文阐述Hps天然免疫和适应性免疫机制的研究现状,以及恩诺沙星对Hps感染保护性免疫应答的潜在影响。

Isolation and Identification of Canine Enterococcus faecalis

With the development of the urban pet industry,suppurative diseases of companion animals are increasing day by day.To explore its associated pathogens,this study was conducted on etilolgical identification from a case of a canine uterus with pus.Bacteriological detection methods were used to isolate and purify the collected disease materials,such as smear staining,microscopic examination and biochemical identification.Then,16S rRNA molecular sequence analysis was used to identify the isolate.It was found that the morphological features of the isolated strain and the biochemical results were consistent with Enterococcus faecalis.The 16S rRNA of the isolate was also as high as 99% homologous with the reference stains.Drug susceptibility test showed that the isolate was resistant to tetracycline,penicillin G,minocycline,erythromycin,ciprofloxacin,norfloxacin,vancomycin,levofloxacin,clindamycin,polymyxin B,chloramphenicol,compound sulfamethoxazole,and clarithromycin,and only susceptible to oxacillin and nitrofurantoin.These results provided reference for the treatment of this type of multidrug-resistant enterococcal infection.

Genetic Evolution Analysis of Actinobacillus pleuropneumoniae

In order to explore the genetic evolution of Actinobacillus pleuropneumoniae(App) in different countries and clarify the relationships among different App in each region, the 16 S rRNA gene of App in the NCBI nucleotide database was analyzed and compared by the bioinformatics method. The phylogenetic tree was constructed after tailoring alignment. The results showed that a stable genetic phenomenon was indicated in the evolutionary process of App. The isolates derived from China were clustered and showed a high degree of conservation. They had a certain genetic relationship with the British and American strains, but had far relationship with the strains from Japan which was a neighboring country of China. The isolates from different countries in the Eurasian continent shared high homology. The isolates of the two regions originated from common ancestors.

Study on Formation and Inhibition Mechanism of Biofilm of Pig Actinobacillus pleuropneumoniae

Pig Actinobacillus pleuropneumoniae(App) could induce chronic respiratory tract infection in pigs, which causes major economic losses on pig industry. Bacterial biofilm(BBF) is bacterial community adsorbed on the surface of biomaterials or body cavity, to protect bacteria escape, and recurrent outbreaks of related infectious diseases and chronic infections resulting therefrom are called bacterial biofilm diseases. App BF belongs to polymers with spatial structure in vitro, and its formation is regulated by multiple genes. Among them, gene deletion of the key component TolC of multidrug efflux pumps and type I secretion systems causes that App BF adhesion weakens; gene deletion of catalytic core ClpP of Clp proteolytic complex induces the inhibition of BF formation; outer membrane lipoprotein VacJ of App promotes BF formation; gene deletion of active enzyme LuxS enhances the formation of App BF and decreases bacterial adhesion ability; gene deletion of Adh obviously declines bacterial accumulation, BF formation and adhesion to host cells. In this paper, BF formation or inhibition mechanism in App is elaborated from molecular level, which could provide reference basis for exploring the prevention of its biofilm diseases.

天津部分地区奶牛乳房炎细菌学分析被引量:1

乳房炎是奶牛业最常见且损失最为严重的疾病之一。为了解天津地区奶牛场乳房炎病原,本研究对天津地区4个奶牛场的77份乳样进行了病原菌的分离与鉴定分析。通过病原分离、细菌形态学观察、生化试验鉴定发现,临床型及亚临床型乳房炎乳样分离菌均以金黄色葡萄球菌和大肠杆菌为优势菌群。药敏试验结果表明,分离菌对环丙沙星、头孢他啶、氧氟沙星、复方新诺明、庆大霉素、卡那霉素、四环素和新霉素敏感,对其他抗菌素药物均呈现不同程度的耐药。本研究明确了天津地区奶牛乳房炎的主要病原,并筛选出了敏感药物,这对该病的防治具有指导意义。

铜绿假单胞菌遗传演化分析被引量:1

为研究铜绿假单胞菌(Pseudomonas aeruginosa,Pa)的遗传演化规律,对2018年本实验室分离出两株铜绿假单胞菌,经基因测序得到16s rRNA基因序列,利用NCBI中的BLAST对比搜索与分离菌株具有相近基因序列的假单胞菌株,采用lasergene中Megalign软件构建基因进化树并比较分析。结果表明:Pa菌株在基因进化树上聚为一支,说明Pa进化较稳定,分离出的两株Pa与来自日本的Pa(登录号为NR_113599)关系最为亲近,他们聚在一个次级分支上;分离菌株与来自美国的两株Pa(登录号为NR_117678与NR_114471)关系较亲近,而与来自德国的Pa(登录号为NR_026078)关系则较远。
