The relationship between SPARC expression in primary tumor and metastatic lymph node of resected pancreatic cancer patients and patients' survival被引量:5

BACKGROUND: Previous researches in pancreatic cancer demonstrated a negative correlation between secreted protein acidic and rich in cysteine (SPARC) expression in primary tumor and survival, but not for SPARC expression in lymph node. In the present study, we aimed to evaluate the SPARC expression in various types of tissues and its impact on patients' prognosis. METHODS: The expression of SPARC was examined by immunohistochemistry in resected pancreatic cancer specimens. Kaplan-Meier analyses and Cox proportional hazards regres- sion were applied to assess the mortality risk. RESULTS: A total of 222 tissue samples from 73 patients were collected to evaluate the SPARC expression, which included 73 paired primary tumor and adjacent normal tissues, 38 paired metastatic and normal lymph nodes. The proportion of posi tive SPARC expression in metastatic lymph node was high (32/38), whereas in normal lymph node it was negative (0/38). Positive SPARC expression in primary tumor cells was associ- ated with a significantly decreased overall survival (P=0.007) and disease-free survival (P=0.003), whereas in other types of tissues it did not show a predictive role for prognosis. Univariate and multivariate analyses both confirmed this significance. CONCLUSION: SPARC can serve a dual function role as both predictor for prognosis and potentially biomarker for lymph node metastasis in resected pancreatic cancer patients.

胺基化碳纳米管基因载体转染细胞的研究

目的 探讨胺基化碳纳米管携带质粒转染细胞并获得稳定转染的可行性,同时验证其生物安全性.方法 胺基化碳纳米管携带质粒pcDNA-GFP-N1转染中国仓鼠卵巢(CHO)细胞,48 h后荧光显微镜下确认转染成功,加入G418(ll00 mg/L)筛选稳定转染细胞;采用XTT法测定胺基化碳纳米管的细胞毒性.结果 胺基化碳纳米管携带质粒成功转染CHO细胞;通过G418筛选2周,最终获得稳定转染细胞.胺基化碳纳米管无明显的细胞毒性.结论 胺基化碳纳米管可以携带外源基因进入细胞,并且可以将其整合到宿主的染色体进行稳定转染.胺基化碳纳米管的生物安全性较好.

LyP-1-MWNTs—GEM的制备及其体外靶向抑制人胰腺癌细胞增殖效应的研究被引量:3

目的观察LyP-1靶向多壁碳纳米管(LyP-1-MWNTs)携载吉西他滨(GEM)对人胰腺癌细胞的体外杀伤效应。方法共价合成法制备LyP-1MWNTs,采用噻唑蓝(MTF)法检测0.1、1.0、10.0、25.0、50.0、75.0mg/L浓度LyP-1-MWNTs—GEM对淋巴结高转移特性胰腺癌细胞株BxPC-3-LN5作用24、48、72h后生长抑制效应,流式细胞仪检测细胞凋亡率。结果LyP-1-MWNTs—GEM对细胞增殖抑制作用呈浓度和时间依赖性,在50.0、75.0mg/L浓度时,LyP-1-MWNTs—GEM组抑制率均显著高于MWNTs-GEM和GEM组(P〈0.05);LyP-1-MWNTs—GEM组抑制率时间相关性规律为:72h〉48h〉24h(P〈0.05,除外75.0mg/L时72h比48h)。流式细胞仪测定LyP-1-MWNTs—GEM组细胞凋亡率(28.68%)亦高于MWNTs.GEM(19.79%)和GEM(16.73%)组。结论LyP-1-MWNTs-GEM在一定浓度时能依赖靶向效能显著抑制BxPC-3-LN5细胞增殖,促进其凋亡。
