光系统ⅡD1蛋白表达调控机制研究进展被引量:3

强光下植物光系统Ⅱ(PSⅡ)易于失活,D1蛋白是PSⅡ受到伤害的原初部位,导致psbA基因编码的D1蛋白降解,同时在多步PSⅡ修复循环中D1蛋白重新合成。衣藻(Chlamydomonas reinhardtii)psbA基因的表达在mRNA加工过程、翻译起始水平上受到严格调控。光通过激活结合在psbA基因mR-NA5’非翻译区(5’-UTR)上的蛋白复合体(RNA-结合蛋白,RBP)而促进翻译,该蛋白质复合体含有氧化还原激活的调节位点。对PSⅡD1蛋白的转化过程、psbA基因转录及翻译进行分析,综述了光系统Ⅱ蛋白表达调节机制的研究进展。

Relationships between Phosphatidylglycerol Molecular Species of Thylakoid Membrane Lipids and Sensitivities to Chilling-induced Photoinhibition in Rice被引量:6

In an attempt to explore the relationships between phosphatidylglycerol (PG) molecular species of thylakoid membrane lipids and sensitivities to chilling-induced photoinhibition, PG molecular species, D1 protein, electron transport activities of thylakoid membrane and the potential quantum yield (FvlFm) in rice treated under middle and low photon flux density (PFD) at 11℃ were analyzed by high performance liquid chromatography, enzyme hydrolysis, gas phase chromatography (GC) and so on. Results showed that the major molecular species of PGs in rice thylakoid membrane were 18:3/16:0, 18:3/16:1(3t), 18:2/16:0, 18:2/16:1(3t), 18:1/16:0, 18:1/16:1(3t), 16:0/16:0, 16:0/16:1(3t). There were large differences in the contents of unsaturated PG molecular species such as 18:1-3/16:0-16:1(3t) and saturated PG molecular species like 16:0/16:0-16:1(3t) among japonica cv 9516 0-9516), japonica-indica hybrid F1 j-9516/i-SY63 (ji-95SY) and indica cv Shanyou 63 (i-SY63). J-9516 containing higher contents of unsaturated PG molecular species was manifest in stable D1 protein contents under chill and tolerant to chill-induced photoinhibition. In contrast to j-9516, i-SY63 with lower contents of unsaturated PG molecular species, exhibited unstable D1 protein contents under chill and was sensitive to chill-induced photoinhibition, ji-95SY containing middle contents of unsaturated PG molecular species between those of j-9516 and i-SY63, exhibited mid extent of sensitivity to chill-induced photoinhibition. The losses in D1 protein also account for the inhibition in electron transport activity of thylakoid membrane and the observed decline in FvlFm. The PG molecular species that is efficient in raising chilling-resistant capacity were those containing unsaturated fatty acids, namely, unsaturated PG molecular species. Substrate Selectivity of Glycerol-3-phosphate Acyl Transferase in Rice被引量:5

Substrate selectivity of glycerol-3-phosphate acyltransferase (EC 2. 3. 1. 15) of rice (Oryza sativa L.) was explored in a comparative study of acyltransferases from seven plant species. In vitro labeling of acyl carrier protein (ACP) with ^14C or 3H showed that acyltransferase from chill-sensitive plants, such as rice that uses either oleic (18:1) or palmitic acid (16:0) as acyl donor at comparable rates, displays lower selectivity than the enzyme from chill-resistant plants, such as spinach, which preferentially uses oleic acid (18:1) rather than palmitic acid (16:0) as an acyl donor. This may be a result of the size and character of the substrate-binding pocket of acyltransferase. Homology modeling and protein structure-based sequence alignment of acyltransferases revealed that proteins from either chill-sensitive or chill-tolerant plants shared a highly conserved domain containing the proposed substrate-binding pocket. However, the aligned residues surrounding the substrate-binding pocket are highly heterogeneous and may have an influence mainly on the size of the substrate binding pockets of acyltransferases. The substrate selectivity of acyltransferase of rice can be improved by enlarging the substrate-binding pocket using molecular biological methods.

Changes in Unsaturated Levels of Fatty Acids in Thylakoid PSII Membrane Lipids During Chilling-induced Resistance in Rice被引量:6

Temperature is one of the abiotic factors limiting growth and productivity of plants. In the present work, the effect of low non-freezing temperature, as an inducer of "chilling resistance", was studied in three cultivars of rice (Oryza sativa L.), japonica cv. 9516 (j-9516), the two parental lines of superhigh-yield hybrid rice between subspecies, Peiai/E32 (ji-PE), and the traditional indica hybrid rice Shanyou 63 (i-SY63). Leaves of chill-treated rice showed chilling-induced resistance, as an increase of their low-temperature tolerance was measured using chlorophyll fluorescence measurements, revealing a change in photosystem II (PSII) efficiency. After 5 d of exposure to 11 ~C under low light (100 pmol·m^-2·s^-1), levels of unsaturated fatty acids in PSII thylakoid membrane lipids decreased during the initial 1-2 d, then increased slowly and reached 99.2%, 95.3% and 90.1% of the initial value (0 d) in j-9516, ji-PE and i-SY63, respectively, on the fifth day. However, under medium light (600 μmol·m^-2·s^-1), all cultivars experienced similar substantial photoinhibition, which approached steady state levels after a decline in levels of unsaturated fatty acids in PSII thylakoid membrane lipids to about 57.1%, 53.8% and 44.5% of the initial values (0 d) in j-9516, ji-PE and i-SY63 on the fifth day. Under either chilling-induced resistance (the former) or low temperature photoinhibition (the latter) conditions, the changes of other physiological parameters such as D1 protein contents, electron transport activities of PSII (ETA), Fv/Fm, xanthophyl cycle activities expressed by DES (deepoxide state) were consistent with that of levels of unsaturated fatty acids in PSII thylakoid membrane lipids. So there were negative correlations between saturated levels of fatty acids (16:1(3t), 16:0, 18:0), especially the 16:1(3t) fatty acid on thylakoid membrane and other physiological parameters, such as D1 protein contents, ETA and (A+Z)/(

紫黄质循环是紫黄质(V)经过中间物单环氧玉米黄质(A)形成玉米黄质(Z)的可逆转换,是光合系统聚光复合体在低光下的聚光状态与高光下的能量耗散状态之间的转换开关。叶黄素中的玉米黄质可钝化(去激发)激发三线态叶绿素(3 Chl*)和激发单线态氧(1 O2*),紫黄质循环可直接或间接地通过非光化学淬灭(NPQ)耗散PSⅡ天线蛋白中的过量光能。天线蛋白被认为是依赖玉米黄质(Z)耗散过量光能的部位,天线蛋白通过结合紫黄质循环组分(V,A和Z)来调节紫黄质循环。类囊体膜脂的性质和结构影响紫黄质循环组分(V,A和Z)间的转换,V的脱环氧化速率依赖于V在类囊体膜脂上侧向扩散的速率,紫黄质脱环氧化作用第一步(由V到A的转换)的速度常数是第二步(由A到Z的转换)速度常数的4~6倍。现有的结果表明,天线蛋白和类囊体膜脂是紫黄质循环最基本的调节器。该文对近年来国内外关于紫黄质循环的基本反应及其功能、紫黄质循环酶结构性质和辅因子以及天线蛋白和类囊体膜脂对紫黄质循环的调节作用及其机理等方面的研究进展进行了综述。

高盐低温胁迫下水稻叶细胞ROS清除系统的相关基因表达被引量:8

采用Affymetrix水稻表达谱芯片,分析高盐和低温胁迫下水稻叶细胞内ROS清除系统的相关基因表达状况,探讨在响应非生物胁迫过程中水稻植株体内抗氧化防卫体系的积极作用。结果表明:(1)水稻叶细胞内ROS清除系统涉及187个基因和/或EST,由抗氧化的非酶类物质如抗坏血酸、谷胱甘肽、生育酚等和抗氧化酶如超氧化物歧化酶(SOD)、抗坏血酸过氧化物酶(APX)、过氧化氢酶(CAT)等组成。(2)在低温逆境下,籼稻(i-93-11)叶细胞表达上调基因(2倍以上,下同)有5个,下调(0.5倍以下,下同)基因有2个;粳稻(j-NJ-1)叶细胞表达上调基因5个,下调基因6个。(3)在高盐胁迫下,籼稻(i-93-11)叶细胞表达上调基因有31个、下调基因13个;粳稻(j-NJ-1)叶细胞表达上调基因27个,下调基因25个。(4)高盐和低温胁迫下水稻叶细胞ROS清除系统相关基因的表达在籼粳稻间存在较大差异,并根据水稻基因表达谱芯片数据构建了水稻响应高盐和低温胁迫ROS清除网络图。

为了阐明籼稻、粳稻对低温强光敏感性的差异 ,着重研究了低温强光下水稻类囊体膜脂不饱和度与紫黄质脱环氧化酶 (VDE)活性的变化。随着低温强光处理时间的延长 ,类囊体膜脂不饱和脂肪酸含量降低、饱和脂肪酸含量增加 ,因而膜脂不饱和指数 (IUFA)下降。同时 ,叶黄素循环的关键酶———VDE活性降低 ,叶黄素循环组分中紫黄质 (V)含量增加 ,而单环氧玉米黄质 (A)和玉米黄质 (Z)的含量减少 ,表现为 (A +Z) / (A +Z +V)比值下降。Arrhenius分析证明 ,VDE对低温和膜脂不饱和度都敏感。相关分析表明 ,类囊体IUFA分别与VDE活性、(A +Z) / (A +Z +V)和D1 蛋白量呈显著的正相关。与粳稻 95 16相比 ,籼稻汕优 6 3类囊体膜的IUFA较低 ,低温下类囊体膜脂流动性和稳定性较差 ,VDE活性和 (A +Z) / (A +Z+V)比值较低。
